Postdoc will participate in the lecture series “Mitochondrial integrated physiology“.
Aims of studies:
To study role of mitochondrial matrix deacetylase Sirtuin 3 in relation to the state of oxidative phosphorylation in normal, diabetic, and cancer cells, and in relations to mitochondrial DNA genetics and morphology of mitochondrial network. Proteomics of protein posttranslational modifications will be studied as well as under conditions of silencing of the key proteins contributing to the cell redox homeostasis (such as IDH2, ALR, mito transhydrogenase, NOX4, etc.) in conjunction with respirometry, confocal fluorescence microscopy monitoring of reactive oxygen species, etc. Also metabolic mapping using the 13C labeled compounds will be employed. The morphology will be studied using the 3D superresolution microscopy (a prototype of BiplaneFPALM instrument is available) in relation to stress stimuli and pathologies, including to situations of mitochondria-specific autophagy, mitophagy.
